In this investigation 100 indigenous Ghezel sheep breed male lambs in 4 to 6 months age were used that were randomly chosen from five different flocks in all over the East Azerbaijan province, In these lambs, the preliminarily nucleotide polymorphisms were identified using the PCR-RFLP technique and the relationship between these polymorphisms and fecal egg counts were investigated and also in present study four samples of PCR product randomly selected from each five flock's lambs (one sample from each flock) and were sequenced subsequently. The nucleotide sequencing results of the PCR products were aligned with other registered nucleotide sequence in NCBI (National Center for Biotechnology Information) gene bank for second exon of Ovar-DRB1 gene in various sheep breeds of world. In the results of alignments, nine novel polymorphisms (two deletions, five substitutions and two insertions) were identified in the second exon of MHC-DRB1 gene sequences of Ghezel sheep breeds. Existence of these nucleotide polymorphisms were caused to substitution of all the amino acid sequence and consequently lead to changes in the structure of foreign peptide binding cleft in the DR heterodimeric proteins in the membrane of antigen presenting cells. Existence of novel polymorphisms in the second exon of Ovar MHC-DRB1 gene has led to changes in protein structure and increased the validity of observed association between identified polymorphism and studied trait.}, keywords = {Novel nucleotide polymorphism,Ovar MHC-DRB1 gene,Gastrointestinal nematodes,Iranian indigenous Ghezel shee}, title_fa = {آنالیز بیوانفورماتیکی و شناسایی پلی مورفیسم های جدید نوکلئوتیدی اگزون شماره ی 2 ژن Ovar MHC-DRB1 در گوسفندان نژاد بومی قزل}, abstract_fa = {هدف از این مطالعه شناسایی پلی ­مورفیسم ­های جدید نوکلئوتیدی در اگزون شماره­ ی 2 ژن MHC-DRB1 است که با ایجاد مقاومت ژنتیکی نسبت به نماتد­های دستگاه گوارشی در گوسفندان نژاد بومی قزل ایرانی در ارتباط است. در این تحقیق از 100 بره­ ی نر 4 تا 6 ماهه نژاد بومی قزل استفاده شده بود که به ­طور­تصادفی از میان 5 گله مردمی در سطح استان آذربایجان­ شرقی انتخاب شده بودند و با استفاده از تکنیک PCR-RFLP پلی­ مورفیسم­ های نوکلئوتیدی مقدماتی و ارتباط آن­ها با تعداد تخم­ انگل شمارش شده در مدفوع، بررسی شد. متعاقباً، 4 نمونه محصول PCR از بره ­های هر گله (یک نمونه از هرگله) به­ طور تصادفی انتخاب و توالی ­یابی شدند. نتایج حاصل از توالی­ یابی با سایر توالی­ های نوکلئوتیدی ثبت شده در بانک ژنNCBI  برای نژادهای مختلف از سراسر جهان، مقایسه شدند. در نتایج حاصل از بررسی هم ­ردیفی­ ها، 9 پلی ­مورفیسم جدید (2­جهش حذفی، 5­ جهش جایگزینی و 2­جهش اضافه شدن نوکلئوتید) درتوالی­ های اگزون شماره ­ی 2 ژن MHC-DRB1 در نژاد قزل شناسایی شد. وجود این پلی ­مورفیسم­ های نوکلئوتیدی جدید باعث تغییر توالی ­های آمینو­اسیدی شده و در نتیجه منجر به ایجاد تغییرات در ساختار شکاف اتصال پپتیدهای بیگانه در پروتئین هترودایمریک DR موجود در غشاء سلول ­های ارائه کننده ­ی آنتی­ ژن ­ها می ­شود به­ طور­کلی احتمالاً وجود پلی­ مورفیسم ­های جدید در اگزون شماره­ ی­2 این ژن باعث تغییر ساختار پروتئین مربوط به آن شده و این موضوع نیز باعث افزایش اعتبار ارتباط بین پلی­ مورفیسم شناسایی شده و صفت مورد بررسی می­ شود.}, keywords_fa = {پلی مورفیسم نوکلئوتیدی جدید,ژن MHC-DRB1,نماتدهای دستگاه گوارش و گوسفند نژاد بومی قزل ایرانی}, url = {https://bilj.asnrukh.ac.ir/article_144030.html}, eprint = {https://bilj.asnrukh.ac.ir/article_144030_d59ca71735729ad52ab5244422670ced.pdf} }
